General description

Serpin A3 is also known as α-1-antichymotrypsin (ACT or AACT) gene with five exons and four introns spanning 12.8kb long, is mapped to human chromosome 14q32.13. The encoded protein is a 423 amino acid with the molecular weight of ~ 55-66kDa and it belongs to the family of serpin superfamily of protease inhibitors. Serpin A3 consists of three β sheets, eight α helices and an active site situated within a hypervariable reactive center loop (RCL).

Biochem/physiol Actions

Overexpression of SERPIN A3 leads to endometrial carcinoma (EC), Therefore targeting SERPIN A3 can be a potent therapeutic method for EC. In addition, elevated expression of SERPIN A3 expression has been observed in various types of carcinomas including breast carcinoma, hepatocellular carcinoma, prostate carcinoma and adenocarcinoma of the lung. SERPINA3 is associated with the pathogenesis of chronic obstructive pulmonary disease (COPD), Parkinson′s disease (PD), Alzheimer′s disease (AD), Stroke, Cystic Fibrosis, Cerebral Haemorrhage and Multiple System Atrophy. SERPIN A3 inhibits the activity of pancreatic chymotrypsin, leukocyte, cathepsin G, mast cell chymases, human glandular, kallikrein 2, kallikrein 3 (prostate specific antigen), pancreatic cationic elastase and an uncharacterized lung serum protease. SERPIN A3 can bind to DNA, this property is unique among the members of SERPIN family.

Physical form

Lyophilized from 0.22 μm filtered solution in 20 mM HEPES, 150 mM NaCl, pH 7.5. Normally Mannitol or Trehalose is added as protectants before lyophilization.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in sterile PBS, pH 7.4 to a concentration of 50 μg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 month. For extended storage, it is recommended to store at -20°C.
