General description

Activity-regulated cytoskeleton-associated protein (Arc) is a growth factor and immediate-early gene (IEG) that is enriched in the brain. Arc is located in the distal dendrites and spine. The Arc gene is located on the human chromosome at 8q24.3.

Specificity

Anti-Arc(N-terminal) specifically recognizes humanand rat Arc.

Immunogen

synthetic peptide corresponding to a sequence at the N-terminal region of human Arc, conjugated to KLH. The corresponding sequence is highly conserved (90% sequence identity) in rat and mouse Arc.

Application

Anti-Arc (N-terminal) antibody produced in rabbit may be used in:
  • immunoblotting
  • immunoprecipitation
  • immunohistochemistry

Biochem/physiol Actions

Activity-regulated cytoskeleton-associated protein (Arc) induction is required for late long-term potentiation (LTP) and memory consolidation. It also plays a role in LTP-induced cofilin phosphorylation and F-actin stabilization. This protein affects synaptic plasticity. Arc regulates the endocytosis of α-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid (AMPA) receptors through its interaction with endocytic proteins endophilin2/3 and dynamin. It is associated with notch signaling and spine morphology.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution bycentrifugation before use. Working dilutions should be discarded if not used within 12 hours.
