General description

ColorWheelColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheelColorWheel® antibody with any ColorWheelColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheelColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheelColorWheel® technology requires both ColorWheelColorWheel® antibody and ColorWheelColorWheel® dye for Flow Cytometry application.

Flow Cytometry Analysis:

Staining of one million HeLa cells was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1114, Anti-Human CD55 (143-30) ColorWheelColorWheel® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheelColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of Cat. No. CWA-1013 Mouse IgG1 isotype control and Cat. No. CWDS-PE ColorWheelColorWheel® Antibody-Ready Phycoerythrin (PE) Dye.

Tested Applications

Target description

Complement decay-accelerating factor (UniProt: P08174; also known as CD55) is encoded by the CD55 (also known as CR, DAF) gene (Gene ID: 1604) in human. CD55 is a phosphatidyl-inositol anchored glycoprotein that inhibits autologous complement cascade activation. It is expressed on the plasma membranes of all cell types that are in contact with plasma complement proteins. It is also found on the surfaces of epithelial cells lining extracellular compartments. Its variants are present in body fluids and in extracellular matrix. Seven different isoforms of CD55 have been described that are produced by alternative splicing. Isoforms 1 and 2 are membrane associated and isoforms 3 to 7 are secreted proteins. The major form of CD55 is a monomer; however, a minor non-disulfide-linked, covalent homodimeric form has also been reported. It is synthesized with a signal peptide (aa 1-34) and a propeptide (aa 354-381) that are subsequently cleaved off to generate the mature form (aa 35-353). It has four Sushi domains, and the first Sushi domain (SCR1) is not essential for its function, but SCR2 and 4 are required for the proper conformation for the active site on SCR3. CD55 recognizes C4b and C3b fragments that condense with cell-surface hydroxyl or amino groups when nascent C4b and C3b are locally generated during C4 and C3 activation. Its Interaction with cell-associated C4b and C3b polypeptides interferes with their ability to catalyze the conversion of C2 and factor B to enzymatically active C2a and Bb. This prevents the formation of C4b2a and C3bBb, the amplification convertases of the complement cascade. It also Inhibits complement activation by destabilizing and preventing the formation of C3 and C5 convertases, which prevents complement damage. Mutations in CD55 gene are known to cause complement hyperactivation and angiopathic thrombosis. Overexpression of CD55 has been reported in cases of HT29 colon carcinoma cells. Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
