General description

Protein eyes shut homolog (UniProt Q5T1H1; also known as EGF-like protein 10, EGF-like protein 11, Epidermal growth factor-like protein 10, Epidermal growth factor-like protein 11, Protein spacemaker homolog) is encoded by the EYS (also known as C6orf178, C6orf179, C6orf180, EGFL10, EGFL11, SPAM) gene (ORF UNQ9424/PRO34591, Gene ID 346007) in human. Drosophila eyes shut protein is expressed in photoreceptor cells, the central nervous system, and peripheral sensory ganglia, and is essential for the formation of a matrix-filled intra-rhabdomeral space within the fly′s retina ommatidia, the loss of spacemaker (spam) converts an open rhabdom system to a closed one. Human EYS/SPAM gene likewise is found to be expressed in the eye and the spinal cord, and mutations in human EYS result in retinitis pigmentosa. Human EYS protein contains 27 EGF-like repeats and five laminin G-like (LG1-5) domains (between EGF repeats 21 & 21, 21 & 22, 23 & 24, 25 & 26, and after repeat EGF repeat 27). The presence of the EGF-like repeats is indicative of a role in the Notch signaling pathway, while EYS LG1-5 bear resemblance to the three agrin laminin G domains that are known to interact with several proteins, including α-dystroglycan at the muscle surface.

Specificity

Expected to react with spliced isoforms 1 and 3, but not isoforms 2 and 4, reported by UniProt (Q5T1H1).

Immunogen

Epitope: EGF-like domains 9-11.
GST-tagged recombinant human EYS fragment corresponding to the EGF-like domains 9-11.

Application

Anti-EYS Antibody, clone 3G10.1 is an antibody against EYS for use in Western Blotting, Immunohistochemistry (Paraffin).
Immunohistochemistry Analysis: A 1:250-1,000 dilution from a representative lot detected EYS in human and rat retina tissue sections.
