General description

Inflammation and immune cell response are key to maintaining homeostasis. Inflammation plays a central role in the etiology of many immunologically related diseases, such as infectious disease, osteoarthritis and respiratory disease, as well as neurologic, metabolic and cardiovascular disease and even cancer. However, until now the ability to measure levels of cytokines and chemokines in bovine samples has been limited primarily to ELISAs. Researchers can now measure multiple inflammatory biomarkers simultaneously using our MILLIPLEX® Bovine Cytokine/Chemokine Panel 1, based on the Luminex® xMAP technology. The MILLIPLEX® Bovine Cytokine/Chemokine Magnetic Bead Panel 1 is a 15-plex kit to be used for the simultaneous quantification of any or all of the following analytes in serum, plasma, and tissue culture supernatant samples: IFNγ, IL-1α, IL-1β, IL-4, IL-6, IL-8 (CXCL8), IL-10, IL-17A, IL-36RA (IL-1F5), IP-10 (CXCL10), MCP-1 (CCL2), MIP-1α (CCL3), MIP-1β (CCL4), TNFα, VEGF-A. MILLIPLEX® offers the broadest selection of analytes across a wide range of disease states and species. Once the analytes of interest have been identified, you can rely on the quality that we build into each kit to produce results you can trust. In addition to the assay characteristics listed in the protocol, other performance criteria evaluated during the validation process include: cross-reactivity, dilution linearity, kit stability, and sample behavior (e.g. detectability and stability). Each panel and kit meets stringent manufacturing criteria to ensure batch-to-batch reproducibility. Each MILLIPLEX® panel and kit includes:Quality controls (QCs) provided to qualify assay performanceComparison of standard (calibrator) and QC lots to a reference lot to ensure lot-to-lot consistencyOptimized serum matrix to mimic native analyte environmentDetection antibody cocktails designed to yield consistent analyte profiles within panelThe MILLIPLEX® Bovine Cytokine/Chemokine Magnetic Bead Panel 1 is part of the most versatile system available for cytokine and chemokine research. From our single to multiplex biomarker solutions, we partner with you to design, develop, analytically validate and build the most comprehensive library available for protein detection and quantitation.

Specificity

Cross-reactivity between the antibodies and any of the other analytes in this panel is non-detectable or negligible.

Application

The Bovine Cytokine/Chemokine bead-based multiplex cytokine panel, using the Luminex® xMAP technology, enables the simultaneous analysis of 15 cytokine and chemokine biomarkers in bovine serum, plasma and cell culture samples. This is an overnight (16-18 hours at 2-8 °C) or two-hour (20-25°C) incubation assay; although an overnight incubation is recommended.A maximum of 25 μL per well 1:2 diluted serum or plasma can be used. Tissue culture or other media may be run neat or appropriately diluted in control medium. This kit may be used for the analysis of all or any combination of the Bovine Cytokine/Chemokine Panel 1 analytes in serum, plasma or tissue/cell/tumor culture supernatant.Analytes available:IFNγ;IL-1α;IL-1β;IL-4;IL-6;IL-8;IL-10;IL-17A;MIP-1α;IL-36RA;IP-10;MCP-1;MIP-1β;TNFα;VEGF-A
