Quality Control Testing
Evaluated by Western Blotting in SH-SY5Y cell lysate.
Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected FRS2/SNT-1 in SH-SY5Y cell lysate.
Tested Applications
Flow Cytometry Analysis: 1 µg from a representative lot detected FRS2/SNT-1 in one million SH-SY5Y cells.
Affinity Binding Assay: A representative lot of this antibody bound FRS2/SNT-1 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected FRS2/SNT-1 in human kidney tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected FRS2/SNT-1 in A-431 cells.

Fibroblast growth factor receptor substrate 2 (UniProt: Q8WU20; also known as FGFR substrate 2, FGFR-signaling adaptor SNT, Suc1-associated neurotrophic factor target 1 (SNT-1)) is encoded by the FRS2 (also known as ) gene (Gene ID: 10818) in human. Fibroblast Growth Factor Receptor Substrate 2 (FRS2) is an adaptor protein characterized by a myristoylated N-terminal region, which facilitates its membrane localization, and a phosphotyrosine-binding (PTB) domain that anchors it to the fibroblast growth factor receptor (FGFR). The C-terminal region of FRS2 contains multiple tyrosine residues that can be phosphorylated, forming docking sites for downstream signaling molecules such as Grb2 and SHP2. FRS2 plays a crucial role in mediating signal transduction from the FGFR to downstream pathways, primarily the MAPK/ERK and PI3K/AKT pathways, which are essential for cellular differentiation, growth, and survival. Upon FGFR activation, FRS2 becomes phosphorylated, serving as a scaffold to recruit and activate various signaling complexes. This activation is pivotal in translating extracellular growth factor signals into cellular responses. Overexpression or hyperactivation of FRS2 can lead to enhanced signaling through pathways that drive tumorigenesis.
外觀
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
