Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Ubiquilin 4 in Raji and HepG2 cell nuclear extract.
Affinity Binding Assay: A representative lot of this antibody bound Ubiquilin 4 peptide with a KD of 4.0 x 10-7 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Ubiquilin 4 in MCF-7 cells.

Ubiquilin-4 (UniProt: Q9NRR5; also known as Ataxin-1 interacting ubiquitin-like protein, A1Up, Ataxin-1 ubiquitin-like-interacting protein, A1U, Connexin43-interacting protein of 75 kDa, CIP75) is encoded by the UBQLN4 (also known as C1orf6, CIP75, UBIN) gene (Gene ID: 56893) in human. Ubiquilin-4, a member of the UbL-UBA protein family, serves as an adaptor in the degradation of specific substrates via the proteasomal pathway. It is highly expressed in pancreas, kidney, skeletal muscle, heart, and throughout the brain. Its levels are reported to be upregulated in aggressive tumors. It serves as a regulator of protein degradation and mediates the proteasomal targeting of misfolded, mislocalized or accumulated proteins. It acts by binding polyubiquitin chains of target proteins via its UBA domain and by interacting with subunits of the proteasome via its ubiquitin-like domain. Ubiquilin-4 colocalizes with the proteasome, both in nucleus and cytoplasm and is exported from the nucleus following interaction with DESI1/POST. In response to DNA damage and phosphorylation at serine 318 by ATM, it localizes to the nucleus and is recruited to sites of DNA damage where it represses homologous recombination repair. It acts by binding and removing ubiquitinated MRE11 from damaged chromatin, leading to MRE11 degradation by the proteasome. MRE11 degradation then prevents homologous recombination repair and redirects double-strand break repair toward non-homologous end joining. Mutations in UBQLN4 gene are shown to cause Amyotrophic lateral sclerosis (ALS). Defects in UBQLN4 gene also cause intellectual impairment, growth retardation, microcephaly, and facial dysmorphism. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
