植物亲水蛋白抽提试剂盒(Plant Hydrophilic Protein Extraction)是专为植物生命科学应用而设计的产品,能够从任意植物种类或组织中抽提亲水蛋白的定性样品。本试剂盒包含两类试剂:一份植物特异性的蛋白酶抑制剂cocktail、一份用于抽提亲水蛋白质的专用配方试剂(植物蛋白抽提试剂第1类试剂,Plant Protein Extraction Reagent Type 1)。蛋白酶抑制剂cocktail是作用于丝氨酸、半胱氨酸、金属蛋白酶、天冬氨酸与氨肽酶等酶类的多种蛋白酶的广谱抑制剂混合物。本品含有胃酶抑素A,E-64,亮肽素,4-(2-氨基乙基)苯磺酰氟(AEBSF)中,1,10-菲咯啉,和苯丁抑制素等多种成分。经验证,cocktail抑制剂混合物能够在抽提过程中高效避免蛋白降解的发生。

在去除了多酚,鞣质等干扰物质之后,新鲜或冻存的植物组织经研磨后,用一个专用配方试剂重悬,来抽提亲水蛋白。通过离心沉淀植物碎片,并收集蛋白抽提液。最后用户可获得水溶性的蛋白样品,用于下游蛋白组学分析。
