1, 4-哌嗪二乙磺酸(PIPES)是一种两性离子缓冲液,属于乙磺酸缓冲液系列,由Good等人率先推出,是一种用于细胞培养、电子显微镜组和其他生物学应用织固定的无毒缓冲液。PIPES 不与金属离子相互作用。由于其在37°C时的pKa接近生理pH,因此在生物、生化和环境研究中被用作缓冲物质。PIPES满足特定的标准:中值pKa,最大水溶性且在所有其他溶剂中的溶解度极小,盐效应极低,pKa随温度变化极小,化学和酶学稳定,在可见光或紫外光谱范围吸光度极小,易于合成。可将碱溶液添加到不含 PIPES 的酸中并滴定至适当的 pH 值,或通过混合单钠盐和二钠盐的等摩尔溶液并滴定至适当的 pH 值,制备缓冲液。

應用

PIPES 钠盐缓冲液可用于:

  • 配制 BRB80 缓冲液,以通过荧光显微镜对动态微管和单个驱动蛋白分子成像
