甘油,通常也是指丙三醇,是一种3-碳糖醇,形成脂肪酸骨架。它是所有脂类合成的中心成分,是甘油三酯和磷脂的骨架,在细胞膜结构中起着重要作用。凭借低毒性,甘油被广泛用于制药、食品和化妆品配方,是通过酯交换法生产生物柴油的主要废物副产物。 除了试剂盒外,当需要进行少量反应时,可分别购买单独的试剂和甘油标准品。
應用
游离甘油测定试剂盒使用偶联酶反应测定游离内源性甘油,不包括初始脂肪酶水解。 甘油被5′-三磷酸腺苷(ATP)磷酸化,从而通过甘油激酶(GK)催化反应形成1-磷酸甘油(G-1-P)和5′-二磷酸腺苷(ADP)。 G-1-P随后被磷酸甘油氧化酶(GPO)氧化为磷酸二羟基丙酮(DAP)和过氧化氢(H2O2)。 过氧化物酶催化H2O2 与4-氨基安替比林(4-AAP)和n -乙基- n -(3-磺丙基)间苯胺钠(ESPA)偶联得到醌亚胺染料,其吸光度在540 nm处最大。540 nm处的吸光度增量与样品中的游离甘油浓度成正比。
游离甘油测定试剂盒用于测定体外培养的哺乳动物细胞释放到培养基中的甘油水平。
甘油三酯和自由甘油试剂盒可以用来定量测试甘油,总甘油三酯或自由甘油三酯。
