转染操作可将小干扰RNA(siRNA)直接递送至细胞中。X-tremeGENE siRNA转染试剂是一种脂质体转染试剂,为向动物细胞中递送siRNA提供了一种方便、可靠和高效的途径,便于用户进行基因敲除后的细胞及功能学研究。
这一创新型试剂能够与siRNA以及siRNA和质粒的混合物(共转染的情况下)形成复合物,并有效递送这些核酸进入细胞,以介导基因沉默。只需简单的几步就可完成转染:首先将转染试剂与稀释过的siRNA混合并孵育,之后将复合物直接加入细胞培养物中。由于X-tremeGENE siRNA转染试剂在有无血清的条件下均工作良好,且经论证无明显细胞毒性,因此无需更换培养基。本品不含任何动物源性成份。

组分

本试剂经0.2 μm滤膜过滤,聚丙烯小管包装。

應用

X-tremeGENE siRNA 转染试剂能够有效将短干扰RNA(siRNAs)转入许多种常用细胞,包括HeLa,NIH 3T3,HEK-293,CHO-K1,和COS-7和一些难转染细胞系,比如HT29,一种人腺癌细胞系。

品質

活性分析:将X-tremeGENE siRNA转染试剂(1 - 2.5 μl)与特异性靶向HPRT看家基因的siRNA(0.1 - 0.35 μg)相互混合。在含10%胎牛血清(FBS)的条件下,将此混合物用于转染HEK-293细胞(生长密度为30-50%的单层细胞)。转染后通过LightCycler®实时荧光定量PCR来检测细胞中HPRT mRNA水平的降低情况。测得的mRNA数据显示,观察到的敲除效率通常处于70-95%。

细胞毒性分析:HEK-293细胞暴露于siRNA/X-tremeGENE siRNA试剂混合物中72小时,未对含有血清的培养基进行换液。之后通过WST-1细胞增殖试剂(Roche)分析细胞,以测试细胞毒性。
