αα-半乳糖基神经酰胺(KRN7000)是恒定自然杀伤T细胞(iNKT)的高亲和性糖脂配体。具有1个α-半乳糖基神经酰胺核心和1个agelasphin-9b神经酰胺核心变体。它是来自海绵的半乳糖神经酰胺(α‐GalCer)的合成同系物。由长链脂肪酰(C26)和鞘氨醇碱基(C18)组成。
應用
αα-半乳糖基神经酰胺(KRN7000)已用于自然杀伤细胞的体外刺激。适合用作骨髓来源细胞(BMDC)和原代肝细胞的脂质抗原,以便进行抗原提呈试验。也适合作为配体激活恒定自然杀伤T细胞(iNKT)。
生化/生理作用
αα-半乳糖基神经酰胺(KRN7000)给药导致辅助性T细胞(TH0)或TH2细胞因子应答。在各种体外模型中表现强效的抗肿瘤活性,比如B6黑色素瘤、结肠腺癌Colon26和肝转移瘤。
