Specificity

Clone 2C14 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Interleukin-4. It targets an epitope within 25 amino acids from the C-terminal part.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal part of human Interleukin-4.

Application

Quality Control Testing

Evaluated by Western Blotting with human recombinant Interleukin-4.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected human recombinant Interleukin-4 .

Tested Applications

Affinity Binding Assay Analysis: The binding affinity of this antibody to Interleukin-4 peptide was at least one thousand-fold (1,000X) higher than that obtained with the non-specific control peptide.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Interleukin-4 in MOLT-4 cells with and without treatment with TPA (40 nM, 6 hrs) and Ionomycin (2 uM 6 hrs) and then treated with Brefeldin A (1ug/mL) for the last 5 .

Enzyme Immunoassay (ELISA): A representative lot of this antibody detected Interleukin-4 protein in ELISA application.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Interleukin-4 (UniProt: P05112; also known as B-cell stimulatory factor 1 (BSF-1), Binetrakin, Lymphocyte stimulatory factor 1, Pitrakinra) is encoded by the IL4 gene (Gene ID: 3565) in human. Interleukin-4 is a pleiotropic cytokine with a well-defined helical bundle structure. It plays a crucial role in regulating immune responses, particularly in promoting the differentiation of naive T cells into Th2 cells and stimulating B cell growth and antibody production. Interleukin-4 is primarily produced by activated T cells, basophils, and mast cells, and its expression can be induced by various stimuli, including antigens, T cell receptor signaling, and cytokines such as IL-2 and IL-4 itself. Upon secretion, Interleukin-4 binds to its receptor, initiating signaling cascades that activate the JAK-STAT pathway and modulate gene expression. Dysregulation of Interleukin-4 has been linked to allergic diseases, such as asthma and atopic dermatitis, as well as autoimmune disorders and certain cancers, highlighting its significance as a therapeutic target in these conditions. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
