General description

Protein disulfide isomerase family A, member 3 (PDIA3) is a liver microsomal protein with molecular mass of 58kDa. It is associated with the protein disulfide isomerase activity. Mature protein consists of two thioredoxin-like active site motifs (CGHC), a nuclear localization motif (KPKKKKK) at its C-terminus end and an endoplasmic reticulum-retention/retrieval motif (QEDL). It has been found in the lumen of the endoplasmic reticulum network, which is a major source for secreting disulphide-bonded polypeptides.

Application

Biochem/physiol Actions

Along with the isomerase activity PDIA3 (protein disulfide isomerase family A member 3) is also involved in the carnitine acyltransferase or protease activities. It participates in different cellular functional activities: (1) hydroxylation of proline residues in procollagen, (2) disulphide bond oxidation, isomerization and reduction, (3) a major non-nuclear binding protein of the thyroid hormone 3,3′,5-triiodo-L-thyronine and (4) a component of the oligosaccharide transferase. It also acts as a chaperone, a subunit of prolyl 4-hydroxylase and microsomal triglyceride transfer protein.
