Specificity

Clone E06 is a mouse monoclonal antibody that binds to oxidized phospholipids and reacts with oxidized LDL, oxidized HDL and proteins covalently modified by oxidized phospholipids.

Immunogen

B lymphocytes from the spleens of ApoE-deficient mice fused with a myeloma cell line and selected hybridomas were cloned and cells were injected intraperitoneally into mice as described by Hörkkö et al. (1999).

Target description

Several reactive lipid peroxidation products are generated during the oxidation of LDL, and modification of lipids and apoproteins occurs in vivo. These oxidized and/modified LDL have the ability to bind to scavenger receptors that can lead to unregulated accumulation of cholesterol in macrophages and formation of foam cells. These products can also react with lipids, apoproteins, and proteins and generate immunogenic neoepitopes and contribute to atherogenesis. The lipid peroxidation products can for adducts with free amino groups of lysines and other amino acids in apolipoproteins. Reactive oxygen species (ROS) and oxidized phospholipids are also reported to activate TRPA1 and TRPV1 cation ion channels that shape thermal and mechanical hypersensitivity in inflammation and pain. These channels are also suggested to be involved in pathological events, including rheumatoid arthritis and trigeminal neuropathic pain. Even locally injected oxidized lipids can elicit thermal and mechanical hypersensitivity and acute nociception. Clone E06/T15 is a monoclonal autoantibody that specifically binds to the head group of oxidized phosphatidylcholines in oxidized phospholipids. (Ref.: Mohammadi, M., et al. (2018). Br. J. Physical form

Purified mouse monoclonal antibody IgM in PBS without preservatives.

Reconstitution

0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
