Specificity

Clone 1G17 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects GATOR complex protein DEPDC5. It targets an epitope within 19 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal half of human GATOR complex protein DEPDC5.

Application

Quality Control Testing

Evaluated by Immunocytochemistry in K562 cells.

Immunocytochemistry Analysis: A 1:100 dilution of this antibody detected DEPDC5 in K562 cells.

Tested Applications

Flow Cytometry Analysis: 0.1 µg from a representative lot detected DEPDC5 in K562 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected DEPDC5 in human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound DEPDC5 peptide with a KD of 1.4 x 10-6 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

GATOR complex protein DEPDC5 (UniProt: O75140; also known as DEP domain-containing protein 5) is encoded by the DEPDC5 (also known as KIAA0645) gene (Gene ID: 9681) in human. DEPDC5 is a component of the GATOR 1 complex that includes DEPDC5, NPRL2, and NPRL3. Its expression is observed in developing and adult brain. It acts as an inhibitor of the amino acid sensing branch of the TORC1 pathway. It acts as a negative regulator of mTOR signaling. Its DEP domain (Dsh-Egl10-pleckstrin) is localized in amino acids 1187-1262. Human cells heterozygous for DEPDC5 loss-of-function mutations are reported to be haploinsufficient for control of mTORC1 signaling. DEPDC5 +/- neurons exhibit hyperactivation of mTORC1 and enlarged somas, a phenotype that can be rescued with mTOR1 inhibition. Nonsense and missense mutation in DEPDC5 gene are known to cause familial temporal lobe epilepsy that is characterized by focal seizures arising from different cortical regions in different family members. Eight isoforms of DEPDC5 have ben described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
