General description

HiTrap Desalting is a prepacked, ready to use column for group separation between high and low molecular weight substances, i.e., buffer exchange prior to or after different chromatographic steps, removal of low molecular weight contaminants or removal of reagents to terminate a reaction.

Application

Gel filtration has successfully been employed for size based separations of macromolecules since the late 1950s. Gel filtration, or size exclusion chromatography, may be performed in three principally different modes, depending on the size differences of the solutes to be separated (i) group separation (ii) fractionation (iii) determination of molecular mass (distributions) When the size difference is large, i.e. a factor of more than 10, we talk about group separation (e.g. desalting, buffer exchange) and when the size difference is small, i.e. a factor of 2 to 5, we talk about fractionation. The third mode is analytical gel filtration, e.g. determination of molecular mass distributions, where an array of molecular masses is to be separated. These different modes put different requirements on the equipment and media used and will yield widely different productivity in terms of information or material processed.

Features and Benefits

  • Handles sample volumes of 1.5 mL, or larger volumes by connecting up to five columns in series.
  • Replaces dialysis as the method of choice for desalting.
  • Desalting requires less than one minute due to the high rigidity of Sephadex G-25 and the optimized packing technology.

Storage and Stability

Please be aware this product may be shipped 90 days before the expiration date. For more information on the batch specific expiration date, please contact technical service.
Store at 4 to 30 °C (20% Ethanol)

This page shows how to separate IgG antibodies by affinity chromatography using Protein G GraviTrap from Cytiva.

Antibodies are needed for research and industrial purposes in different quantities, from microgram to kilogram scale. It is important to design and use a purification method that will yield protein of a quality and quantity that is adequate for the particular application.

The yield of GST-tagged proteins is highly variable, ranging from 1 mg/l to 10 mg/l. The yield depends on various parameters, such as nature of the tagged protein, the host cell, and the culture conditions used.
