Specificity

Clone 1M15 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Ras-related protein Rab-14. It targets an epitope within 23 amino acids from the C-terminal region.

Immunogen

KLH-conjugated liner peptide corresponding to 23 amino acids from the C-terminal region of human Ras-related protein Rab-14.

Application

Quality Control Testing

Evaluated by Western Blotting in Raji cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Rab14 in Raji cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Rab14 in lysate from Mouse brain tissue and 1:1,000 dilution detected Rab14 in Rat brain tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Rab14 in Human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound Rab14 peptide with a KD of 8.4 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Rab14 in U-251 MG cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Ras-related protein Rab-14 (UniProt: P61106; also known as Rab14) is encoded by the RAB14 gene (Gene ID: 51552) in human. Rab proteins constitute a large family of the low-molecular-weight GTPases that play key roles in the regulation of intracellular membrane trafficking. They also play a role in cargo selection, vesicle budding, vesicle motility, tethering, docking, and fusion. They act as molecular switches that flip between a GDP-bound inactive state and a GTP-bound active form in which they recruit downstream effector proteins onto membranes. Rab14 is involved in membrane trafficking between the Golgi complex and endosomes during early embryonic development. It is reported to regulate the Golgi to endosome transport of FGFR-containing vesicles during early development. It may also act by modulating the kinesin KIF16B-cargo association to endosomes. It is also involved in phagosome and early endosome fusion. It is shown to be essential for the formation of PtdIns(4,5)P2-enriched lipid domains that promote the recruitment of downstream effectors of polarity, including Par3, aPKC and Cdc42. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
