General description

Carbonic anhydrase 4 (CA4) is a 35kDa protein, encoded by the gene mapped to human chromosome 17q23.1-23.2. It is expressed on extracellular surfaces of certain endothelial and epithelial cells. CA4 is one among the 12 isozymes of carbonic anhydrase.
Carbonic anhydrase 4 (CA4) is also known as Carbonate dehydratase IV, Carbonic anhydrase IV, which belongs to the α-carbonic anhydrase family. CA4 may stimulate the sodium/bicarbonate transporter activity of SLC4A4 that acts in pH homeostasis. It is essential for acid overload removal from the retina and retina epithelium, and acid release in the choriocapillaris in the choroid. The enzyme regulation is activated by histamine, L-adrenaline, D-phenylalanine, L- and D-histidine. CA4 can interact with SLC4A4.

Biochem/physiol Actions

Carbonic anhydrase 4 (CA4) plays a vital role in CO2 hydration activity. It also catalyzes the dehydration of HCO3. In addition, it also facilitates CO2 and HCO3 transport in various vital tissues like kidney and lung Polymorphism in the gene has been associated with the development of sporadic retinitis pigmentosa (RP).

Physical form

Lyophilized from 0.22 μm filtered solution in 50 mM Tris, 150 mM NaCl, pH 7.5. Normally Mannitol or Trehalose is added as protectants before lyophilization.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in sterile PBS, pH 7.4 to a concentration of 50 μg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 month. For extended storage, it is recommended to store at -20°C.
