General description

Monoclonal Anti-SORBS1 (mouse IgG1 isotype) is derived from the hybridoma SORBS-437 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide. SORBS1 is an adaptor protein and is mainly expressed in insulin-sensitive tissues such as adipose tissue, liver and skeletal muscle.
Sorbin and SH3 domain-containing protein 1 (SORBS1) is also called c-Cbl-associated protein (CAP) or ponsin. The SORBS1 gene is located on human chromosome 10q24.1. SORBS1 has sorbin homology domain (SOHO) and three SH3 (sarcomahomology 3) domains.

Specificity

Monoclonal Anti-SORBS1 recognizes human, rat and mouse SORBS1.

Immunogen

Synthetic peptide corresponding to an internal sequence of human SORBS1. The isotype is determined by ELISA using Mouse Monoclonal Antibody Isotyping Reagents (Sigma ISO-2).

Application

Monoclonal Anti-SORBS1 antibody produced in mouse may be used in immunoblotting and flow cytometry.

Biochem/physiol Actions

SORBS1 acts as an insulin-signaling molecule by forming a complex with the oncoprotein Cbl.
Sorbin and SH3 domain-containing protein 1 (SORBS1) modulates cell adhesion by interacting with cytoskeletal proteins. It regulates glucose uptake. Polymorphisms in SORBS1 is implicated in obesity, insulin resistance, type 2 diabetes and diabetic nephropathy. It suppresses tumorigenesis by modulating c-Jun N-terminal kinases (JNKs) of mitogen-activated protein kinase (MAPK) signaling pathway in lung and breast cancers.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For extended storage, freeze at -20 0C in working aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
