General description

T-cell surface glycoprotein CD3 zeta chain (CD3ζ), also called CD247, is a component of T-cell antigen receptor (TCR) multimeric complex. CD3ζ comprises three immunoreceptor tyrosine-based activation motifs (ITAMs) with conserved Tyr-X-X-Leu/lle sequence, short ectodomain of nine amino acids and a long cytoplasmic tail region. CD247 gene is mapped to human chromosome 1q24.2 and is majorly expressed in the natural killer and T cells.

Specificity

Anti-phospho-CD3z(pTyr111) recognizes mouse phosphorylatedCD3z(pTyr111).

Immunogen

synthetic phosphopeptide corresponding to residues surrounding Tyr111 of mouse CD3ζ, conjugated to KLH. The corresponding sequence differs by two amino acids in human.

Application

Anti-phospho-CD3ζ (pTyr111) antibody produced in rabbit may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

T-cell surface glycoprotein CD3 zeta chain (CD3ζ orCD3z) signal peptide in the N-terminal region aids in targeting protein translation to the endoplasmic reticulum. It is involved in the activation of the T cells and plays a key role in the recognition of antigenic peptide on antigen-presenting cells or tumor cells. CD3z chain plays a key role in regulating the TCR assembly, transport, and cell surface expression. CD3ζ reorganizes the actin-based cytoskeleton and regulates the morphology of immune cells. Downregulation of CD247also impacts the expression of the zeta chain Of T cell receptor associated protein kinase 70 (ZAP70) of the T-cell signal-transduction pathway. Lack of CD3ζ impairs retinal ganglion cell (RGC) motility and its presence is crucial for synaptic wiring in the retina. The absence of CD3ζ in T cells is postulated to be involved in the pathology of rheumatoid arthritis, systemic lupus erythematosus (SLE) and cancer. It is regarded as a potential therapeutic target for ovarian cancer treatment.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
