General description

Neural precursor cell expressed developmentally down-regulated protein 9 (Nedd9), also known as CRK-associated substrate-related protein (Cas-L) and human enhancer of filamentation 1 (HEF1), is an adhesion adapter protein. Nedd9 gene is mapped to human chromosome 6p24.2. It belongs to a larger family of Cas docking adapter proteins and is expressed preferentially in lymphocytes and epithelial cells.

Biochem/physiol Actions

Neural precursor cell expressed developmentally down-regulated protein 9 (Nedd9) participates in integrin and growth factor signaling pathways that regulate growth, motility and apoptosis. It is thought to play an important role in early neuronal differentiation and brain development. Nedd9 undergoes extensive phosphorylation, a process dependent both on cell adhesion and on the integrity of the actin cytoskeleton. Focal adhesion kinase (FAK) and sarcoma (Src) regulate Nedd9. Nedd9 is implicated in metastatic cancer and is upregulated in solid tumors and ovarian cancer. Overexpression of Nedd9 is implicated in the invasion of glioblastoma and in the pathogenesis of breakpoint cluster region-Abelson (Bcr-Abl)-dependent tumor .

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
