General description

Monoclonal Anti-Chimerin β2 (rat IgG2a isotype) is derived from the hybridoma HAR 2E3 produced by the fusion of mouse myeloma cells and splenocytes from rat immunized with a fusion protein of full length human Chimerin β2. Chimerin β2 (CHN2) is a member of the chimerin family and encodes a protein with a phorbol-ester/ diacylglycerol (DAG)-type zinc finger containing C1 domain, a Rho-GTPase activating proteins (GAP) domain and an Src homology 2 (SH2) domain. Chimerin β2 gene is mapped to human chromosome 7p14.3.

Application

Monoclonal Anti-Chimerin β2 antibody produced in rat has been used in immunoblotting at a dilution of 1:1000 and immunocytochemistry.

Biochem/physiol Actions

Chimerin β2 protein (CHN2) is a high affinity receptor for the phorbol esters and diacylglycerol (DAG). Binding of DAG induces translocation of the protein from the cytosol to the Golgi apparatus membrane. It has guanosine triphosphatase GTPase-activating protein activity regulated by phospholipid binding. Decreased expression of CHN2 gene is associated with high-grade gliomas and breast. Deregulation in expression of CHN2 gene is associated with lymphomas, glioblastoma and insulin resistance. Mutations in the CHN2 gene have been associated with schizophrenia in men.

Physical form

Solution in 0.01M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.

Storage and Stability

Store at -20 °C. For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze at -20 °C in working aliquots. Repeated freezing and thawing,, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
