Specificity
Clone 1F10 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Transcription factor Spi-C. It targets an epitope within 18 amino acids from the N-terminal region.

KLH-conjugated linear peptide corresponding to 18 amino acids from the N-terminal region of human Transcription factor Spi-C.
Application
Quality Control Testing
Evaluated by Western Blotting in Jurkat cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SPI-C in Jurkat cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected SPI-C in Raji cell lysate.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SPI-C in Jurkat cells.
Affinity Binding Assay: A representative lot of this antibody bound SPI-C peptide with a KD of 6.6 x 10-7 in an affinity binding assay.

Target description
Transcription factor Spi-C (UniProt: Q8N5J4; also known as Spi-C) is encoded by the SPIC gene (Gene ID: 121599) in human. Spi-C is an E26 transformation-specific transcription factor that is closely related to PU.1 and Spi-B. It binds to the PU-box, a purine-rich DNA sequence (5′-GAGGA[AT]-3′) that can act as a lymphoid-specific enhancer. Its expression is observed in fetal and adult spleen and lymph nodes and at lower levels in bone marrow and fetal liver. Spi-C plays an important role in antibody-generating responses, B cell development, and red pulp macrophage generation. It is induced by heme- and NF-κB-dependent pathways in macrophages. In the B cell compartment, its activity is tightly regulated during development and differentiation. It is reported to promote the transition from large to small pre-B cells and regulate the generation of antibody-secreting cells. In the myeloid lineage, Spi-C has an crucial role in generating splenic red pulp macrophages through a heme-dependent pathway. It is also involved in regulating the inflammatory profile of macrophages in response to NF-κB signaling where it promotes a protective, anti-inflammatory phenotype. Spi-B and Spi-C are reported to play opposing roles in secondary antibody responses and plasmablast differentiation. They both interact with intronic regions of the Bach2 locus to regulate transcription. However, Spi-B activates Bach2 whereas Spi-C represses Bach2. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
