Specificity

Clone 1G13 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Superoxide Dismutase 1 (SOD1).

Immunogen

His-tagged, full-length (without the initiator methionine), recombinant human Superoxide Dismutase 1 (SOD1).

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected SOD1 in Jurkat cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected SOD1in HepG2 cell lysate.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected SOD1 in Jurkat cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected SOD1 in human liver tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound recombinant Human SOD1 with a KD of 9.2 x 10-6 in an affinity binding assay.

Target description

Superoxide dismutase [Cu-Zn] (UniProt: P00441; also known as EC:1.15.1.1, Superoxide dismutase 1, SOD1) is encoded by the SOD1 gene (Gene ID: 6647) in human. Superoxide dismutases are metalloproteins that dismutate the superoxide radical (O2•) to hydrogen peroxide (H2O2) and molecular oxygen.  Three types of SOD isozymes have been identified in eukaryotic cells. Cu/Zn superoxide dismutase (SOD1) is predominantly located in the cytosolic fractions; Mn superoxide dismutase (SOD2) is located in the mitochondria, and the extracellular superoxide dismutase (SOD3) is localized in the extracellular space. SOD1 is a homodimeric enzyme that contributes to about 90% of the cellular SOD activity. It has a β-barrel structure that is stabilized by a Cys57-Cys146 disulfide bridge and a zinc ion in the active site. The two subunits are held together by strong hydrophobic forces, which makes it one of the most compact and stable protein. Each subunit also contains a copper ion that undergoes alternative oxidation-reduction cycle in the course of the dismutation of superoxide to oxygen and H2O2. Although it is predominantly expressed in the cytoplasm, it is also found in the nucleus and mitochondria. In addition to its antioxidant properties, it is also implicated in signal transduction, and both its wild-type and mutant forms are prone to misfolding. A vast number of mutations have been recorded in SOD1, some of these mutations cause amyotrophic lateral sclerosis (ALS), a neurodegenerative disorder predominantly affecting upper and lower motor neurons. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
