General description

The REDExtract-N-Amp PCR reaction mix is a PCR Master mix containing buffer, salts, dNTPs, and REDTaq DNA polymerase.
This PCR ReadyMix is intended for use with Sigma′s Extract-N-Amp Plant PCR kit and Extract-N-Amp Tissue PCR Kit. All Extract-N-Amp kits are sufficient for one PCR amplification per extraction. However, where additional PCR reactions are required, this supplemental PCR ReadyMix may be needed. The REDExtract-N-Amp PCR ReadyMix contains an inert tracking dye that allows for convenient direct loading of PCR products onto agarose gels for analysis.

Application

REDExtract-N-Amp PCR Master Mix has been used:
  • for the genotyping of fixed zebrafish embryo samples
  • in polymerase chain reaction (PCR) amplification of the DNA isolated from lichen family using the REDExtract-N-Amp Plant PCR Kit
  • in the amplification of DNA isolated from single yeast colonies and fungal spores using REDExtract-N-Amp seed PCR kit.

REDExtract-N-Amp PCR Master Mix is also suitable for colony PCR.

Features and Benefits

  • A Gel-ready- No loading buffer or tracking dye needs to be added. The PCR product is loaded directly into an agarose gel after amplification. The red tracer migrates at approximately the same rate as a 125bp fragment (slightly faster than bromophenol blue).
  • Convenience- Hot-start feature in JumpStart products allows reaction set up at room temperature, which can be beneficial when working with multiple samples. Ready-mix format reduces setup time.
  • Reproducibility- Reduces the risk of contamination from multiple pipet steps and provides consistent performance across both reactions and time.
  • High Specificity & Yield- JumpStart Taq DNA polymerase, an antibody inactivated hot-start enzyme, is designed to minimize non-specific amplification while increasing target yield & specificity.

The Extract-N-Amp™ Tissue PCR Kit has been created to release PCR-ready DNA from mouse tails in a 15 minute single tube procedure. The included 2x PCR mix is optimized to work with the crude extracts and the solutions in the kit

Standard methods for extracting DNA from tissues can be extremely laborious and time consuming. Certain applications, such as genotyping of transgenic mice using a section of tail, employ a lengthy DNA extraction process.

Protocols

The REDExtract-N-Amp Plant PCR kit includes an inert loading dye in the ReadyMix for easy gel loading after amplification. This protocol provides a rapid protocol to extract plant genomic DNA for amplification using the optimized ReadyMix.

The exrract-N-Amp kit protocol provides a rapid DNA extraction method that leads to a PCR-ready sample in 15 minutes. Optimized PCR ReadyMix yields successful and clean amplification. RED loading dye allows for sample tracking.
