R-Spondin-1 (Rspo-1) 属于 Wnt 调节剂 (Rspo) 家族。目前,(Rspo) 家族由 4 个结构相关的分泌型配体 (Rspo 1-4) 组成,均含有弗林蛋白酶样和凝血酶敏感蛋白结构域。Rspo-1 在发育中的中枢神经系统的某些区域,以及肾上腺、卵巢、睾丸、甲状腺和气管中表达。重组人 R-Spondin-1 是由 243 个氨基酸残基组成的 26.7 kDa 蛋白。由于糖基化,在还原条件下,通过 SDS PAGE 分析,反应蛋白-1 以大约 40.0kDa 的表观分子量迁移。
生化/生理作用
Rspo (R-Spondin) 可与 Frizzled/LRP6(LDL 受体相关蛋白 6)受体复合物相互作用,刺激 Wnt/β-连环蛋白信号通路。Rspo-1 基因的破坏引起隐性综合征伴 XX 性反转、掌跖角化过度和易患皮肤鳞状细胞癌。Rspo-1 还与肿瘤细胞的抗辐射能力有关。

从 10 mM 磷酸钠 (pH 7.5) 冻干。
重構
打开前离心小瓶。用水复溶至 0.1-1.0 mg/mL 浓度。切勿涡旋。该溶液可在 2-8°C 条件下至多储存 1 周。为了延长储存时间,建议在含有载体蛋白(例如 0.1%BSA)的缓冲液中进一步稀释,并在-20°C 至-80°C 下储存。
