一般說明

多聚-L-赖氨酸是一种带正电荷的氨基酸聚合物,每个赖氨酸残基约有一个HBr。氢溴酸盐可使聚-L-赖氨酸以可溶于水的结晶形式存在。在β结构中可能会发现少量的产物,因为HBr干扰氨基与羧基或含N或O部分之间的氢键。聚-L-赖氨酸氢溴酸盐是非特异性附着因子,促进细胞附着在基质上。

應用

用0.5 ml的0.1 mg/ml溶液覆盖25cm2时,建议将其作为细胞培养基质。必须对每种细胞系和应用确定最佳附着条件。
已将聚-L-赖氨酸氢溴酸盐用于将皮质组织的解离细胞附着进培养板。 也用于处理玻片,将样本固定其上进行免疫组化分析。

生化/生理作用

多聚-L-赖氨酸可用作包被物质,以促进细胞在培养物表面更好地附着。研究表明多聚-L-赖氨酸(PLL)包被的培养板可使细胞生长速度加快。在PLL包被的底物上培养间充质干细胞(MSC)时,它有助于维持其干性特征(stemness properties)并防止衰老。包被PLL的培养板表现出改善的MSC生长,并使与细胞粘附、分化、增殖和信号传导相关的基因表达上调。此外,PLL包被的多孔表面改善细胞增殖并有助于MSC的成骨分化。
