小鼠单克隆抗荧光素酶抗体可识别转染细胞中的重组荧光素酶。该抗体可为用户提供荧光素酶的一种替代检测方法。由于它可直接检测荧光素酶蛋白,因此不依赖于荧光素酶活性或快速反应动力学的测量。这还使得原位检测荧光素酶的表达成为可能,并在样品组之间具有可重复性。
小鼠单克隆抗荧光素酶抗体(小鼠IgG1同种型)来源于LUC-1 杂交瘤,其通过融合小鼠骨髓瘤细胞和萤火虫荧光素酶免疫的BALB /c小鼠的脾细胞产生。荧光素酶是一种受体蛋白,含有两个结合区,一个为ATP结合区,另一个为荧光素底物结合区。两个结合区彼此非常接近。

特異性

小鼠单克隆抗荧光素酶抗体可识别转染 (293T) 的真核细胞的重组荧光素酶。

應用

小鼠单克隆抗荧光素酶抗体已被用于:

生化/生理作用

荧光素酶在存在底物荧光素以及 Mg2+、氧和ATP的情况下催化生物荧光反应。荧光素酶检测非常迅速、灵敏,而且和CAT检测不同,不需要反射性底物。抗荧光素酶抗体可以原位检测荧光素酶表达,提供了一种研究信号序列激活中细胞定位的方法。

工作稀释度:1-2 μg/ml

儲存和穩定性

如需连续使用,可在2-8 °C保存至多一月。 若需延长储存时间,可将溶液分装并冷冻。 不建议反复冻融或存放在“无霜”冰柜中。 如果长期储存后发生轻微浑浊,请在使用前通过离心澄清溶液。 如果在12小时内未使用,则应丢弃工作稀释样品。
