hCP抗体(又称CALP)可定位哺乳动物平滑肌中的钙调蛋白。在人子宫中,一种约 27 kDa 的附加条带(1-钙调蛋白)也可能随之染色。该抗体不会与骨骼肌、心脏或非肌肉组织的钙调蛋白发生交叉反应。然而,该抗体与人或小鼠骨骼肌中的表位(150 kDa 范围)表现出交叉反应。在免疫组化染色中,该产品表现出平滑肌特异性。它可以染色组织切片和原代培养细胞(或早期传代细胞)中的血管和内脏平滑肌细胞,但不能染色大多数最初源自平滑肌的细胞系。该抗体不能染色上皮、内皮或结缔组织成纤维细胞。 该产品不与鸡肉的平滑肌组织发生交叉反应。

生化/生理作用

钙调蛋白是钙结合蛋白,属于肌动蛋白相关蛋白家族。钙调蛋白是血管平滑肌(VSM)中蛋白激酶 C(PKC)自磷酸化所必需的。钙调蛋白-1 抑制可防止子宫平滑肌细胞迁移、引起形态变化和F-肌动蛋白重排,而不会影响其增殖和凋亡。钙调蛋白 H1(CN)是平滑肌细胞的分化标志物,在几种人类肿瘤的血管中下调。它可以抑制肌动球蛋白 ATP 酶活性。h1 和 h2 钙调蛋白与 F-肌动蛋白结合,并在调节平滑肌和非肌肉细胞的肌动蛋白丝中发挥关键作用。
