General description

Insulin receptor substrate-1 (IRS-1) is among the four IRS proteins (IRS-1-IRS-4) that mediate signalling by insulin receptor (IR) and insulin-like growth factor (IGF) receptor. IRS-1 has a pleckstrin homology domain and a phosphotyrosine-binding domain. . Phosphorylation of Tyr612 and Tyr632 fully activate the PI3K signalling pathway in response to insulin and induce translocation of GLUT4. Thus IRS-1 protein plays a vital role in mediating the metabolic actions of insulin via the PI3K pathway. Additionally, IRS-1 has the abililty to translocate to the nucleus, the mechanism of which is not very clear. The transcriptional activity of IRS-1 is involved in the development of breast and prostate cancers
The antibody recognizes human IRS-1 (pTyr612). It may cross react with in mouse, rat and chicken (100% homologous) IRS-1 (pTyr612).

Immunogen

synthetic phosphopeptide derived from the region of insulin receptor substrate-1 (IRS-1) phosphorylated at tyrosine 612.

Application

Anti-phospho-IRS-1 (pTyr612) antibody may be used for detection by immunoblotting.
Applications in which this antibody has been used successfully, and the associated peer-reviewed papers, are given below.

To examine contributions of specific YXXM motifs in human insulin receptor substrate-1 (IRS-1) to mediating the metabolic actions of insulin, we studied IRS-1 mutants containing various substitutions of Phe for Tyr. In transfected NIH-3T3(IR) cells, insulin stimulation caused a 5-fold

Sulfonylureas (SUs) have been suggested to have an insulin-independent blood glucose-decreasing activity due to an extrapancreatic effect. However, a lack of adequate in vivo evidence makes this statement controversial. Here, we aimed to evaluate whether glimepiride has extrapancreatic blood glucose-lowering

Defects in the gene coding for dysferlin, a membrane-associated protein, affect many tissues, including skeletal muscles, with a resultant myopathy called dysferlinopathy. Dysferlinopathy manifests postgrowth with a progressive loss of skeletal muscle function, early intramyocellular lipid accumulation, and a striking
