Cell Line Description

Established after spontaneous transformation of cells isolated from the lung of a normal Chinese hamster (male). The G1 phase in these cells is either absent or very short. Consequently they have been used in studies of the G1 growth period. A clone called V79.4 is available (Sigma Catalogue number. 93010723). V79 is well established in toxicology studies. Stability of karyotype and morphology is making them suitable for gene toxicity assays with low background aberrations. The cells do not express endogenous cytochrome P450 and have been used for the insertion and expression of P450 from human, rat, mouse and fish.

Application

Growth factors and transformation studies, mutagenesis study
V79 has been used to detect structural and numerical chromosome damage by measuring the formation of micronuclei in interphase cells that have been through a mitotic division.

Culture Medium

DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:12 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
