General description
Guanine nucleotide-binding protein G(q) subunit alpha (UniProt P50148; also known as Guanine nucleotide-binding protein alpha-q) is encoded by the GNAQ (also known as GAQ, SWS) gene (Gene ID 2776) in human. Heterotrimeric G proteins are activated by G protein-coupled receptors (GPCRs) and are composed of alpha (α), beta (β), and gamma (γ) subunits. There exists also monomeric Ras superfamily of small GTPases that are commonly referred to as small G-proteins. G-proteins mediate signal transduction initiated by GPCRs. Ligand binding activates GPCRs by inducing a conformational change. The activated GPCRs catalyzes the GDP-to-GTP exchange of the Gα subunit, causing a dissociation of the Gβγ dimer from the now GTP-bound Gα subunit. The Gα subunit gradually hydrolyzes the bound GTP to GDP and eventually re-associates with Gβγ and re-starts the cycle. There exist at least 20 different Gα subunits, which are separated into four main groups, G1, Gs, Gq, and G12/13. Four different genes code for Gq alpha subunits, GNAQ, GNA11, GNA14, and GNA15. Gq proteins mediate a variety of cellular signaling events by activating phospholipase C (PLC). Gq-coupled GPCRs include α1-adrenoreceptor, angiotensin II receptor type 1, calcitonin receptor, muscarinic receptors (M1, M3, and M5), histamine H1 receptor, Serotonin 5-HT2 receptors, vasopressin type 1 receptors 1A and 1B.
Specificity
Clone 8A11.1 was raised against a human Gqα (GNAQ) recombinant fragment. Clone 8A11.1 reactivity toward Gq/11α (GNA11), Gq/14α (GNA14), and Gq/15α (GNA15) has not been tested.
