Specificity

Clone 3F14 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects C-X-C motif chemokine 10 (CXCL10). It targets an epitope within 16 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the N-terminal half of human C-X-C motif chemokine 10 (CXCL10).

Application

Quality Control Testing

Evaluated by Western Blotting with recombinant Human CXCL10.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected recombinant Human CXCL10.

Tested Applications

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected CXCL10 in human thymus and human tonsil tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CXCL10 in human thymus tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound CXCL10 peptide with a KD of 1.3 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

C-X-C motif chemokine 10 (UniProt: P02778; also known as 10 kDa interferon gamma-induced protein, Gamma-IP10, IP-10, Small-inducible cytokine B10) is encoded by the CXCL10 (also known as INP10, SCYB10) gene (Gene ID: 3627) in human. CXCL10 is a secreted protein that is synthesized with a signal peptide (aa 1-21), which is subsequently cleaved off to produce the mature form. It is mainly secreted by monocytes, endothelial cells as well as fibroblasts and is expressed by epithelial cells in thymus tissue. Microglial cells are also reported to produce CXCL10 in response to viral stimulation. It is a pro-inflammatory cytokine that is involved in a wide variety of processes, including chemotaxis, differentiation, and activation of peripheral immune cells, regulation of cell growth, apoptosis, and modulation of angiostatic effects. It plays an important role in stimulating the activation and migration of immune cells to the site of infection. Binding of CXCL10 to the CXCR3 activates G-protein- mediated signaling and results in downstream activation of phospholipase C-dependent pathways. Activation of the CXCL10/CXCR3 axis also plays an important role in neurons in response to brain injury for activating microglia and directing them to the lesion site. Cleavage of CXCL10 by dipeptidyl peptidase IV (DPP4) removes two amino acids from the N-terminal region that leads to formation of an antagonistic form of CXCL10, a dominant negative form that can bind to CXCR3 but does not induce signaling. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
