General description

Integrin alpha-6 (UniProt P23229; also known as CD49 antigen-like family member F, CD49f, Integrin alpha6B, VLA-6) is encoded by the ITGA6 (also known as ITGA6B) gene (Gene ID 3655) in human. The biological functions attributed to cell-extracellular matrix (ECM) interactions are mediated primarily by integrin family transmembrane receptors. A total of 18 alpha and 8 beta integrins have been identified in human. Each alpha integrin pairs up with one beta integrin non-covalently to form a functional heterodimeric dimer. At least 24 distinct integrin heterodimers are known with varying ligand specificities. For example, the alpha6-beta1 integrin is reported to mediate the spreading and laminin-binding of Schwann cells, while the alpha6-beta4 integrin is shown to play a role in the suppression of anoikis in human intestinal epithelial cells (IECs).

Specificity

Expected to react with Integrin alpha-6 spliced isoforms 2 (Alpha-6X1A), 4 (Alpha-6X2A ), 6 (Alpha-6X1X2A), 7, and 9, as well as their respective proforms, cleaved light chains, and processed integrin alpha-6 forms (Alpha6p). Epitope is not present in isoform 1 (Alpha-6X1X2B), 3 (Alpha-6X1B), or 5 (Alpha-6X2B).

Immunogen

Epitope: Cytoplasmic region of isoforms 2/4/6/7/9
KLH-conjugated linear peptide corresponding to the cytoplasmic region of human Integrin alpha6 isoforms 2/4/6/7/9.

Application

Immunohistochemistry Analysis: A 1:50 dilution from a representative lot detected Integrin alpha6 in human skin tissue.

This Anti-Integrin alpha6 Antibody, clone 6H7.1 is validated for use in Western Blotting and Immunohistochemistry (Paraffin) for the detection of Integrin alpha6.

Quality

Evaluated by Western Blotting in SW480 human colon cancer cell lysate.

Western Blotting Analysis: 1.0 µg/mL of this antibody detected Integrin alpha6 in 10 µg of SW480 human colon cancer cell lysate.

Target description

~140 kDa observed. Due to glycosylation, the target band(s) appears larger than the calculated 106.6 to 124.0 kDa based on the proform sequences of isoforms 2/4/6/7/9. Uncharacterized band(s) may appear in some lysates.
