General description

Ceramides containing 2-hydroxy fatty acid (hFA) are found primarily in the epidermis. 12:0(2R-OH) Ceramide is a 2R hydroxylated lauric acid-containing ceramide.

Biochem/physiol Actions

Hydroxy fatty acid (hFA)-sphingolipids play a specific role in cell signaling. Synthesis of hFA-ceramides requires fatty acid 2-hydroxylase (FA2H). Mutations of this key enzyme are associated with the development of nervous system disorders such as leukodystrophy and spastic paraparesis in humans. hFA-ceramides are essential for the epidermal permeability barrier function. The mechanism of action for an antitumor drug, PM02734 involves hFA-ceramides in the cell membrane.
