General description

Escort II is a unique formulation of a proprietary cationic lipid and the neutral lipid DOPE. This liposome-forming compound is used for transfection of nucleic acids into eukaryotic cells.

Application

Suitable for transient and stable transfection of nucleic acids into cultured eukaryotic cells. Use approximately 25 μl Escort II and 5 μg DNA (in provided unique DNA Dilution buffer) per 6 cm cell culture plate. Protocol optimization provides very efficient transfection. The following cells have been successfully transfected using Escort II:

B16-F0
BHK-21
CHO-K1
COS-1
COS-7
CV-1
D. mel (S2)
F9
HepG2
HEK293
HeLa
HUVEC
K562
L929
MDCK
NIH3T3
P19
PC-12
Vero

Features and Benefits

  • Suitable for stable and transient transfection
  • High efficiency for a wide variety of cell types
  • Compatible with both serum and serum-free transfection protocols
  • Low toxicity

Components

Escort II Transfection Kit (0.75 mL) contains:
0.75 mL Escort II Reagent (L6037)
0.8 mL Hydration Buffer (B1180)
4 mL DNA Dilutent (D6813)

Escort II Transfection Kit (1.5 mL) contains:
1.5 mL Escort II Reagent (L6037)
1.6 mL Hydration Buffer (B1180)
4 mL DNA Dilutent (D6813)

Note the formulation of Escort II is confidential.

Caution

Do not freeze.

Principle

A stable complex is formed when Escort II is mixed with DNA in the absence of serum. The complexes are stable and can be directly added to the cell culture medium, where they fuse with the cell membrane, releasing the DNA into the cytoplasm. Note: complex formation is inhibited by serum, but once stable complexes have formed, the presence of serum is without consequence.
