General description

pRSFDuet-1 is designed for the co-expression of two target proteins. The vector encodes two multiple cloning sites (MCS) each of which is preceded by a T7 promoter, lac operator, and ribosome binding site (RBS). The vector also carries the pRSF1030 replicon (also known as NTP1), LacI gene, and kanamycin resistance marker.

Application

pRSFDuet-1 DNA - Novagen has been used as a vector:
  • to clone the DNA fragments of defensin α 5 (DEFA5) for recombinant expression
  • to clone the coding region of Mad1597–719
  • to clone the globular domain of Bombyx mori Spc24 (73-162aa) with an N-terminal 6X His-tag to generate the B. mori 6xHis-Spc24(73-162)-Spc25(70-211) pRSF-DUET1 bacterial expression constructs

Accurate chromosome segregation requires assembly of the multiprotein kinetochore complex at centromeres. In most eukaryotes, kinetochore assembly is primed by the histone H3 variant CenH3 (also called CENP-A), which physically interacts with components of the inner kinetochore constitutive centromere-associated network
