Specificity

Clone 1B17 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects human IgG1 Fc chain. It targets an epitope within 16 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the internal region of human IgG1 Fc chain.

Application

Quality Control Testing

Evaluated by Western Blotting with Human IgG, Fc fragment.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected Human IgG1 Fc chain.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot did not detect Mouse IgG, Rabbit IgG, and Chicken IgY.

Flow Cytometry Analysis: 1 µg from a representative lot detected IgG1 Fc chain in one million Raji cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected IgG1 Fc chain in Human tonsil tissue sections.

Enzyme Immunoassay (ELISA): Serial dilutions from a representative lot detected Human IgG1 Fc fragment, but did not detect Mouse IgG and Chicken IgY.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Immunoglobulin heavy constant gamma 1 (UniProt: P01857; also known as Ig gamma-1 chain C region, Ig gamma-1 chain C region EU, Ig gamma-1 chain C region KOL, Ig gamma-1 chain C region NIE) is encoded by the IGHG1 gene (Gene ID: 3500) in human. IgG1 Fc belongs to the constant region of immunoglobulin heavy chains. It enables antigen binding activity and immunoglobulin receptor binding activity and acts upstream of or within several processes, including antibody-dependent cellular cytotoxicity, phagocytosis, and positive regulation of immune response. Two isoforms of IgG1 Fc have been described that are produced by alternative splicing. Isoform 1 is a secreted form and isoform 2 is a single-pass membrane protein with one extracellular domain (aa 1-350), a transmembrane domain (aa 351-371), and a cytoplasmic domain (aa 372-399). IgG1 Fc contains three Ig-like domains (aa 6-99; 121-220; and 229-325) that are localized in the extracellular domain. It is reported to mediate IgG effector functions on monocytes triggering antibody-dependent cellular cytotoxicity (ADCC) of virus-infected cells. Its glycosylation on asparagine 180 is shown to be essential for interaction with Fc receptors and ability to activate complement pathway. Over-expression of IGHG1 has been linked to enhanced proliferation of gastric cancer cells via AKT/GSK-3β/β-Catenin pathway. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
