JTV-519 is a potent inhibitor or the ryanodine receptor 2 (RYR2) blocker. RYR2 is a cardiac calcium channel that regulates calcium levels in the sarcoplasmic reticulum. JTV-519 stabilizes RYR2 in the closed state.

Both receptor families are made up of subunits that are among the largest of known proteins and for each the functional channel comprises a tetramer in which six membrane-spanning helices, near the carboxy terminus of each subunit, form a calcium channel.
