General description

BAD (BCL2 associated agonist of cell death) is a proapoptotic protein. It is a 184 kDa protein made of 168 amino acids. It belongs to the Bcl2 family of proteins. The gene encoding it is localized on human chromosome 11q13.

Peptide sequence around aa. 134-138 (S-R-S-A-P), according to the protein BAD.

Biochem/physiol Actions

Phosphorylated BAD (BCL2 associated agonist of cell death) has the ability to interact with 14-3-3 scaffold proteins in the cytoplasm. Dephosphorylated BAD is active and it moves to the outer membrane of mitochondria and make heterodimers with BCL-XL to prevent BCL-XL antiapoptotic activities.

Target description

The protein encoded by BAD gene is a member of the BCL-2 family. BCL-2 family members are known to be regulators of programmed cell death. This protein positively regulates cell apoptosis by forming heterodimers with BCL-xL and BCL-2, and reversing their death repressor activity. Proapoptotic activity of this protein is regulated through its phosphorylation. Protein kinases AKT and MAP kinase, as well as protein phosphatase calcineurin were found to be involved in the regulation of this protein. Alternative splicing of this gene results in two transcript variants which encode the same isoform.
