General description

Acetylcholine is a multifunctional neurochemical with diverse roles in the brain and various organ systems. It acts as a neurotransmitter, facilitating communication between neurons, myocytes, and cells in glandular tissues. The name "acetylcholine" originates from its chemical composition, which is an ester of acetic acid and choline. Tissues that utilize or respond to acetylcholine are termed cholinergic. While it functions as a neurotransmitter throughout the body, acetylcholine is particularly associated with the neuromuscular junction, where it triggers muscle activation by connecting motor neurons in the ventral spinal cord with muscles. Additionally, acetylcholine serves as a neurotransmitter in the autonomic nervous system, enabling communication between preganglionic and postganglionic neurons. It is the final product released by parasympathetic postganglionic neurons and plays vital roles in skeletal muscle movement, regulation of smooth and cardiac muscle, as well as in learning, memory, and mood.

Principle

The Acetylcholine Assay Kit provides a simple, direct and high-throughput assay for measuring acetylcholine in biological samples. In this assay, acetylcholine is hydrolyzed by acetylcholinesterase to choline which is oxidized by choline oxidase tobetaine and H2O2. The resulting H2O2 reacts with a specific dye to form a pink colored product. The color intensity at 570 nm or fluorescence intensity at λEx = 530 nm/λEm = 585 nm is directly proportional to the acetylcholine concentration in the sample.
