General description

Metadherin, or more commonly known as LYRIC, or AEG-1 (Astrocyte elevated gene-1) is a single pass membrane protein that plays a key role in anchorage-independent growth of immortalized melanocytes and astrocytes. Anchorage-independent division is a critical component in tumor cell expansion and metastasis. Similarly, metadherin activates the nuclear factor kappa- B (NF-kappa-B) transcription leading to expression of numerous genes involved in cell growth and expansion, and it is also involved in HIF-1alpha mediated angiogenesis in tumor growth. Finally new research demonstrates that metadherin interacts with RNA-induced silencing complex (RISC) and plays a very important role in RISC and miRNA functions as well. The protein is a key target of research because of its tumor promoting effects in lung, bone and brain cancers. Metadherin is localized to the endoplasmic reticulum and nuclear membranes in normal tissue, thus staining is perinuclear primarily but Metadherin staining can also be seen in the nucleus, at tight junctions and even at the cell surface in certain cancers. Metadherin expression is activated by TNF, and metadherin is expressed in many cells types with the highest levels in muscle-dominating organs such as skeletal muscle, heart, tongue and small intestine and in endocrine glands such as thyroid and adrenal gland. Metadherin is often over expressed in various cancers including breast, brain, prostate, and melanoma and glioblastoma multiforme. EMD-Millipore’s Anti-metadherin mouse monoclonal, clone 2F11C3 has been tested by western blot against several human cancer cell lysates, by flow cytometry against human HeLa cells, and in human paraffin-embedded human liver tissue.

Application

Detect Metadherin using this mouse monoclonal antibody, Anti-Metadherin Antibody, clone 2F11C3 validated for use in western blotting, IHC & Flow Cytometry.
Immunohistochemistry Analysis: A 1:1,000 dilution from a representative lot detected Metadherin in human breast cancer tissue.

Flow Cytometry Analysis: A 1:200-400 dilution from a representative lot detected Metadherin in HeLa cells

Optimal working dilutions must be determined by end user.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
