General description

IgM exists as a pentamer and is secreted by peritoneal B cells. IgM is a highly conserved antibody in vertebrates and is expressed early during immune response.
IgM is a glycoprotein antibody that regulates humoral immune responses. Mouse IgM is involved in modulating B cell memory. This antibody isotype has also been implicated in the development of autoimmune responses associated with the pathogenesis of type 1 diabetes in mice.
Goat anti-mouse IgM (μ-chain specific)-agarose antibody is specific for mouse IgM when tested against purified mouse IgA, IgG (all subclasses), and IgM myeloma proteins.

Application

Anti-Mouse IgM (μ-chain specific)−Agarose antibody produced in goat has been used in agarose binding assay in primary splenic cells and in immunoprecipitation of soluble lens protein and myelogenous leukemia cell lines.
Applications in which this antibody has been used successfully, and the associated peer-reviewed papers, are given below.
Chromatin immunoprecipitation (1 paper)
Goat anti-mouse IgM (μ-chain specific)-agarose antibody has been used for immunoprecipitation and ChIP assays. The product can also be used for Ouchterlony double diffusion applications.
To isolate chimeric proteins that were fused with the IgM heavy chain, CHO supernatant was ran over a column packed with agarose-linked goat anti-mouse IgM mu chain specific.

Biochem/physiol Actions

IgM displays polyreactive and autoreactive functionality. It plays a key role in tissue homeostasis by mediating clearance of tissue based molecules. IgM has sites for N-linked glycosylation, with sugars namely, mannose, galactose, N-acetyl glucosamine and sialic acid IgM plays a key role in the engulfing of apoptotic cells. A reduction in serum IgM levels leads to increased autoimmune response and higher risk for infections. IgM linked agarose resin have been tested for efficient conjugate binding.
