Target description

LIM/homeobox protein Lhx5 (UniProt: P61376; also known as LIM homeobox protein 5, Homeobox protein LIM-2) is encoded by the Lhx5 (also known as Lim-2, Lim2) gene (Gene ID: 124451) in rat. Lhx5 is a nuclear protein that belongs to a family of proteins that carry the LIM domain. Its LIM zinc-binding domains are localized in amino acids 3-61 and 62-125. It acts as a transcriptional regulator and is reported to play an essential role in the regulation of neuronal differentiation and migration during development of the central nervous system. Its expression is detected in developing Purkinje cells soon after they exited the cell cycle and migrate out of the cerebellar ventricular zone. Lhx5 is also required for the generation of the mamillary body (MM) and its derived tracts that are related to memory and spatial navigation that interconnect hippocampal, thalamic, and tegmental regions. It is predicted to enable DNA-binding transcription factor activity, RNA polymerase II-specific and RNA polymerase II transcription regulatory region sequence-specific DNA binding activity. Lhx5 is reported to regulate the local inhibition of Wnt proteins. Activation of Lhx5 is shown to expand forebrain structures. It can rescue forebrain deficiencies caused by excess Wnt activity. Its inhibition results in ectopic activation of Wnt signaling. It regulates the expression of two secreted Frizzled-related Wnt antagonists, Sfrp1a and Sfrp5 that can reduce the ectopic activation of Wnt signaling and rescue the forebrain deficiencies caused by inhibition of Lhx5 function.
