Gene Information

Specificity

Recognizes a full-length P2X4 purinergic receptor protein and not with other known proteins tested so far.

SPECIES REACTIVITIES: The immunogen sequence is identical in mouse. Other species have not been tested.

Immunogen

A purified peptide of rat P2X4 protein (amino acids 370-388) (Accession P51577).

Application

Anti-P2X4 Receptor Antibody detects level of P2X4 Receptor & has been published & validated for use in WB.

Western blot: 1-1.5 μg/mL (1:200:1:300) using ECL

Immunohistochemistry on rat brain sections.

Dilutions should be made using a carrier protein such as BSA (1-3%)

Optimal working dilutions must be determined by the end user.

Physical form

Affinity purified immunoglobulin. Lyophilized from phosphate buffered saline, pH 7.4, containing 1% BSA, and 0.05% sodium azide as a preservative. Reconstitute with 50 μL of sterile deionized water. Centrifuge antibody preparation before use (10,000 xg for 5 min).

Storage and Stability

Maintain lyophilized material at -20°C for up to 12 months after date of receipt. After reconstitution maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

Analysis Note

Control
CONTROL ANTIGEN: Included free of charge with the antibody is 40 μg of control antigen (lyophilized powder). The stock solution of the antigen can be made up using 100 μL of sterile deionized water. For negative control, preincubate 1 μg of purified peptide with 1 μg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.

We have cloned P2X4, a member of the P2-purinoceptor family, which has a new pharmacological profile. Rat P2X4 is distantly related to P2X1, P2X2 and P2X3 and is expressed in brain, spinal cord, lung, thymus, bladder, adrenal, testis and vas

A cDNA was cloned which encodes a new ATP-gated ion channel (P2X4 receptor). ATP induces a cationic current in HEK293 cells transfected with the P2X4 receptor. However, the current is almost completely insensitive to antagonists effective at other P2X receptors.

There is strong evidence that ATP acts as an excitatory neurotransmitter in the periphery, yet little is known about fast central ATP-mediated transmission. We report here the molecular cloning of a novel neuronal ionotropic ATP receptor of the P2x subtype
