Specificity

Clone 1E6 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects CD16a. It targets an epitope within 25 amino acids from the cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the cytoplasmic domain of human CD16a.

Application

Quality Control Testing

Evaluated by Western Blotting in Human spleen tissue lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CD16a in Human spleen tissue lysate.

Tested Applications

Affinity Binding Assay; A representative lot of this antibody bound CD16a peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD16 in Jurkat cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CD16 in Human spleen tissue sections.

Flow Cytometry Analysis: 1 µg from a representative lot detected CD16 in one million Human peripheral blood mononuclear cells (PBMC) co-stained with PE-conjugated CD56.

Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Low Affinity immunoglobulin gamma Fc region receptor III-A (UniProt: P08637; also known as CD16a antigen, Fc-gamma RIII-alpha, Fc-gamma RIII, Fc-gamma RIIIa, FcRIII, FcRIIIa, FcR-10, IgG Fc receptor III-2, CD16a) is encoded by the FCGR3A (also known as CD16A, FCG3, FCGR3, IGFR3) gene (Gene ID: 2214) in human. CD16a is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-16), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 17-208), a transmembrane domain (aa 209-229), and a cytoplasmic domain (aa 230-254). It serves as a receptor for the invariable Fc fragment of immunoglobulin γ. It does not bind free monomeric IgG, thus avoiding inappropriate effector cell activation in the absence of antigenic trigger. It contains high mannose- and complex-type oligosaccharides and its glycosylation at Asn-180 is shown to be essential for high Affinity binding to the Fc and for discrimination between fucosylated and afucosylated IgG glycoforms. It is expressed on natural killer cells, macrophages, subpopulation of T-cells, immature thymocytes, and placental trophoblasts. CD16 serves as a receptor for the Fc region of IgG and binds complexed or aggregated IgG and also monomeric IgG. Phenylalanine 203 in CD16a determines the transmembrane domains and serine 203 in CD16b determines the GPI-anchoring. CD16 is reported to mediate antibody-dependent cellular cytotoxicity (ADCC) and other antibody-dependent responses, such as phagocytosis and cytokine production. Mutations in FCGR3A gene have been linked to immunodeficiency 20 that is characterized by functional deficiency of NK cells and makes subjects vulnerable to severe herpes viral infections. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
