Specificity

Clone 2D20 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects GRP75. It targets an epitope within 20 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the N-terminal half of human GRP75.

Application

Quality Control Testing

Evaluated by Western Blotting in PC12 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected GRP75 in PC12 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected GRP75 in COS-7, MCF-7, and NIH3T3 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected GRP75 in human kidney tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected GRP75 in MCF-7 cells.

Affinity Binding Assay: A representative lot of this antibody bound GRP75 peptide with a KD of 2.3 x 10-7 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Stress-70 protein, mitochondrial (UniProt: P38646; also known as 75 kDa glucose-regulated protein, GRP-75, Heat shock 70 kDa protein 9, Mortalin, MOT, Peptide-binding protein 74, PBP74) is encoded by the HSPA9 (also known as GRP75, HSPA9B, mt-HSP70) gene (Gene ID: 3313) in human. GRP75 is a mitochondrial resident protein that is synthesized with transit peptide (aa 1-46). It associates with the mitochondrial contact sites and cristae organizing system (MICOS) complex. It is a multipotent chaperone that regulates multiple cellular processes ranging from stress response to neurodegeneration. It plays a key role in several mitochondrial processes, including mitochondrial biogenesis, protein import, folding, and degradation. It has multiple binding partners, including p53, FGF-1, IL-1, IL-1R, HSP60, GRP94, and TIM and TOM. When associated with its binding partners, GRP75 has also reported to have various extramitochondrial functions, such as cell cycle arrest, apoptosis, senescence, and tumorigenesis. Dysregulation of GRP75-induced mitochondrial and proteasomal dysfunction has been suggested to be a contributor to the pathogenesis of Parkinson, Alzheimer s disease, diabetes mellitus, and cardiovascular diseases. Its levels are upregulated in multiple cancer types. Its interaction with p53 in the cytoplasm of transformed cells results in cytoplasmic retention and transcriptional inactivation of p53 leading to tumor growth. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
