Specificity

Clone 2J4 is a ZooMAb rabbit recombinant monoclonal antibody that detects both isoforms of human Phosphoglycerate mutase family member 5 (PGAM5). It targets an epitope within 86 amino acids from the N-terminal half.

Immunogen

His-tagged recombinant fragment corresponding to 86 amino acids from the N-terminal half of human Phosphoglycerate mutase family member 5 (PGAM5).

Application

Anti-PGAM5, clone 2J4 ZooMAb, Cat. No. ZRB1055, is a Rabbit monoclonal antibody that specifically targets PGAM5 and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PGAM5 in lysates from HEK293 cells, mouse testis, and human brain tissue.

Immunohistochemistry (Paraffin) Analysis: 1:100 dilution from a representative lot detected PGAM5 in human testis and human cerebral cortex tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound human recombinant PGAM5 with a KD of 3.0 x 10-9 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PGAM5 in MCF7 cells.

Target description

Serine/threonine-protein phosphatase PGAM5, mitochondrial (UniProt: Q96HS1; also known as EC:3.1.3.16, Bcl-XL-binding protein v68, Phosphoglycerate mutase family member 5) is encoded by the pgam5 GENE (Gene ID: 192111) in human. PGAM5 is a single-pass mitochondrial membrane protein that displays phosphatase activity for serine/threonine residues and is shown to dephosphorylate and activate MAP3K5 kinase. It does not display any phosphoglycerate mutase activity. PGAM5 usually exists as a dimer, but can also form ternary complex with Nuclear Factor, Erythroid 2 Like 2 (NFE2L2) and Kelch Like ECH Associated Protein 1 (KEAP1). Its transmembrane domain is localized to amino acids 7 to 29. A 35 amino acid sequence at its N-terminal function as a non-cleaved mitochondrial targeting sequence that targets the protein to the cytosolic side of the outer mitochondrial membrane. Two isoforms of PGAM5 have been described that are produced by alternative splicing. Both isoforms are reported to be phosphorylated by RIPK1/RIPK3 complex under conditions of necrosis and this phosphorylation is shown to increase PGAM5 phosphatase activity. Overexpression of isoform 1 results in clustering of mitochondria around the nucleus whereas isoform 2 overexpression results in the formation of disconnected punctuate mitochondria distributed throughout the cytoplasm. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

0.3 mg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
