General description

Mouse Sphingosine kinase 2 ( GenBank Accession No. NM_203280), full length with N-terminal His tag, MW = 68 kDa, expressed in a Baculovirus infected Sf9 cell expression system.

Application

Sphingosine kinase 2 is useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Sphingosine kinase 2 has been used in a study to assess blood–brain barrier protection in hypoxic preconditioning-induced cerebral ischemic tolerance. Sphingosine kinase 2 has also been used in a study to investigate sensitization of human colon cancer cells to sodium butyrate-induced apoptosis.

Unit Definition

One unit of sphingosine kinase activity is defined as the amount of enzyme required to produce 1 pmol of S1P/min

Physical form

Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 50% glycerol and 3 mM DTT.

Preparation Note

Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.

Sphingosine kinase (SphK) has emerged as an attractive target for cancer therapeutics due to its role in cell survival. SphK phosphorylates sphingosine to form sphingosine 1-phosphate (S1P), which has been implicated in cancer growth and survival. SphK exists as two

Sphingosine kinases (SphKs) have been recognized as important proteins regulating cell proliferation and apoptosis. Of the two isoforms of SphK (SphK1 and SphK2), little is known about the functions of SphK2. Sodium butyrate (NaBT) has been established as a promising

Protection of the blood-brain barrier (BBB) is correlated with improved outcome in stroke. Sphingosine kinase (SphK)-directed production of sphingosine-1-phosphate, which we previously documented as being vital to preconditioning-induced stroke protection, mediates peripheral vascular integrity via junctional protein regulation.
